The number of registered members would not be a problem in itself. A more critical factor would be server load, I believe, which would depend on how actively the members use the site. As far as I know, it would be manageable.
As for handling that number of users, new users can be added as a batch upload in a CSV file; I assume this works smoothly. I haven't ever needed to use it myself. After that, how to handle the users would depend on what features of Tiki you want to use and so on. I think the question of what site software to use depends mainly on the tools you need, and user number is a secondary consideration.
